چکیده

Objectives This is a protocol for Cochrane Review (intervention). The objectives are as follows: To assess the effectiveness and safety of brexanolone related neurosteroid GABAA positive allosteric modulators in comparison with any other treatment (pharmacological, psychological, or psychosocial), placebo, usual postnatal depression (PND).

Novel positive allosteric modulators of GABAA receptors with anesthetic activity

GABAA receptors are the main inhibitory neurotransmitter receptors in the brain and are targets for numerous clinically important drugs such as benzodiazepines, anxiolytics and anesthetics. We previously identified novel ligands of the classical benzodiazepine binding pocket in α1β2γ2 GABAA receptors using an experiment-guided virtual screening (EGVS) method. This screen also identified novel l...

The testosterone-derived neurosteroid androstanediol is a positive allosteric modulator of GABAA receptors.

Testosterone modulates seizure susceptibility, but the underlying mechanisms are obscure. Recently, we demonstrated that testosterone affects seizure activity via its conversion to neurosteroids in the brain. Androstanediol (5alpha-androstan-3alpha,17beta-diol) is an endogenous neurosteroid synthesized from testosterone. However, the molecular mechanism underlying the seizure protection activit...

Inhibition of recombinant L-type voltage-gated calcium channels by positive allosteric modulators of GABAA receptors.

Benzodiazepines (BDZs) depress neuronal excitability via positive allosteric modulation of inhibitory GABA(A) receptors (GABA(A)R). BDZs and other positive GABA(A)R modulators, including barbiturates, ethanol, and neurosteroids, can also inhibit L-type voltage-gated calcium channels (L-VGCCs), which could contribute to reduced neuronal excitability. Because neuronal L-VGCC function is up-regula...

Discovery of positive allosteric modulators and silent allosteric modulators of the μ-opioid receptor.

μ-Opioid receptors are among the most studied G protein-coupled receptors because of the therapeutic value of agonists, such as morphine, that are used to treat chronic pain. However, these drugs have significant side effects, such as respiratory suppression, constipation, allodynia, tolerance, and dependence, as well as abuse potential. Efforts to fine tune pain control while alleviating the s...

Are AMPA Receptor Positive Allosteric Modulators Potential Pharmacotherapeutics for Addiction?

Positive allosteric modulators (PAMs) of α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors are a diverse class of compounds that increase fast excitatory transmission in the brain. AMPA PAMs have been shown to facilitate long-term potentiation, strengthen communication between various cortical and subcortical regions, and some of these compounds increase the production and r...
